miR-200a在成釉细胞瘤中的表达及意义

Expression and role of miR-200a in ameloblastoma

摘要: [摘要] 目的:研究miR-200a的表达与人成釉细胞瘤之间的相关性。方法:应用RNAhybrid和miRanda软件预测β-catenin基因可能的靶向miRNA;选择30例人成釉细胞瘤标本和5例正常牙胚标本,应用实时荧光定量PCR方法检测标本中miR-200a的表达情况。结果:RNAhybrid和miRanda软件分别在β-catenin mRNA第68~96碱基和第69~94碱基位置预测到一个miR-200a结合位点。与正常牙胚组织标本相比,miR-200a在人成釉细胞瘤组织标本中的表达存在明显的下调,差异显著(P < 0.05)。结论:miR-200a的表达改变与人成釉细胞瘤相关,miR-200a可能为β-catenin的靶基因之一。

Abstract: [Abstract] Objective:To study the relationship between miR-200a expression and human ameloblastoma. Methods:RNAhybrid and miRanda software were applied to predict the β-catenin targeting miRNAs. 30 cases of human ameloblastoma and 5 cases of normal tooth germ tissues were selected to detect the expression of miR-200a by real-time PCR technology. Results:MiRNA targeting prediction showed that both the 68th to 96th base pairs and the 69th to 94th base pairs in β-catenin mRNA contained a miR-200a binding site. Compared with normal tooth germ tissues, the expression of miR-200a showed an obvious down-regulation in human ameloblastoma tissues, which difference was statistically significant (P < 0.05). Conclusions:There is a relationship between the expression of miR-200a with human ameloblastoma, and miR-200a might be one of the targeting gene of β-catenin.
