两种fimA型牙龈卟啉单胞菌对人脐动脉平滑肌细胞增殖的影响

Effects of two fimA variants of Porphyromonas gingivalis on the function of human umbilical artery smooth muscle cells

摘要: 目的:探究两种fimA型牙龈卟啉单胞菌对人脐动脉平滑肌细胞增殖的影响。方法:采用组织块贴壁法培养人脐动脉平滑肌细胞。在厌氧条件下培养ⅠfimA、ⅣfimA型牙龈卟啉单胞菌,并与人脐动脉平滑肌细胞共同培养;在刺激后2、8、24和48h时,采用CCK-8法检测细胞生长状况。结果:ⅠfimA型牙龈卟啉单胞菌刺激人脐动脉平滑肌细胞后8h,促增殖作用明显,但是24和48h平滑肌细胞增殖受到抑制(P<0.05);ⅣfimA型牙龈卟啉单胞菌对人脐动脉平滑肌细胞没有明显促细胞增殖或细胞毒性作用。结论:ⅠfimA型和ⅣfimA型牙龈卟啉单胞菌对人脐动脉平滑肌细胞的作用存在差异,ⅠfimA型牙龈卟啉单胞菌有明显的促平滑肌细胞增殖和细胞毒性作用。

Abstract: Objective:To explore effects of viability on human umbilical artery smooth muscle cells affected by two different fimAgenetypes of Porphyromonas gingivalis. Methods:The tissue-block attached method was used for primary culture of human umbilical artery smooth muscle cells. Porphyromonas gingivalis was cultured in the anaerobic conditions. Human umbilical artery smooth muscle cells were invaded by fimA genotype Ⅰ and Ⅳstrains respectively for 2, 8, 24 and 48 h. The CCK-8 assay was applied in testing viability of smooth muscle cells. Results: After fimA typeⅠstrains were cultured with vascular smooth muscle cells, smooth muscle cells were significantly increasing at 8 h, but obviously, cytotoxicity appeared at 24 h and 48 h (P<0.05). Cells invaded by fimA type Ⅳ Porphyromonas gingivalis did not have any significant change as compared with that of control group. Conclusions:There are diversities between the two different strains. FimA type Ⅰ strain plays a role in the processes of facilitating proliferation and cytotoxicity of vascular smooth muscle cells.
