TNF-α抑制miR-21影响小鼠骨髓基质干细胞成骨分化

TNF-α inhibits mBMSCs osteoporosis through miR-21

摘要: 目的:探讨肿瘤坏死因子-α(TNF-α)是否能通过微小RNA(miR)-21影响小鼠骨髓基质干细胞(mBMSCs)的成骨分化。方法:利用实时定量RT-PCR检测TNF-α对miR-21的影响。在实时定量RT-PCR、Western blot等技术验证TNF-α对mBMSCs成骨能力的影响后,通过转染技术在含有TNF-α的成骨环境下上调miR-21的表达,进而观察转染后mBMSCs的成骨能力。结果:TNF-α抑制mBMSCs成骨分化,转染miR-21至含有TNF-α的成骨环境下的mBMSCs能部分恢复其成骨分化能力。结论:miR-21介导的成骨分化是TNF-α抑制mBMSCs成骨的途径之一。

Abstract: Objective:To investigate whether or not the TNF-α can affect mouse bone marrow stromal stem cells (mBMSCs)osteoporosis through miR-21.Methods:Real-time RT-PCR was used to detect the levels of miR-21 in mBMSCs osteogenesis on condition of TNF-α. Osteogenic gene and protein was determined by real-time RT-PCR and Western blot analysis, respectively. After upregulation the of miR-21 in the presence of TNF-α, the osteogenic potential of mBMSCs were investigated. Results:TNF-α inhibits mBMSCs osteoporosis and miR-21 can restore the potential of mBMSCs treated with TNF-α. Conclusions:TNF-α inhibits mBMSCs osteoporosis through miR-21.
