TEAD2在口腔鳞癌中的表达及其对细胞生物学行为的影响

Expression of TEAD2 and its effect on cell biological behavior in oral squamous cell carcinoma

摘要: 目的:探究口腔鳞癌(OSCC)组织中转录增强相关结构域2(TEAD2)的表达模式及其对OSCC细胞生物学行为的影响。方法:选取97例OSCC患者肿瘤样本和20例口腔黏膜健康者样本,采用免疫组化(IHC)染色检测样本中TEAD2的表达水平,分析其表达与OSCC患者临床参数及预后之间的关系。使用小干扰RNA(siRNA)转染沉默CAL27细胞TEAD2,通过细胞流式实验、划痕实验及Transwell侵袭实验分别检测TEAD2沉默后对CAL27细胞凋亡、迁移及侵袭的影响,通过Western blot和实时荧光定量PCR检测上皮-间充质转化相关蛋白和基因的表达。结果:IHC结果显示,TEAD2在OSCC中表达明显高于正常口腔黏膜(P<0.05)。TEAD2高表达水平与患者肿瘤大小、病理分级、颈部淋巴结转移相关,TEAD2高表达组患者总体及无病生存率低(P<0.05)。TEAD2沉默后,CAL27细胞凋亡能力提高,侵袭、迁移能力明显减弱(P<0.01),E钙黏蛋白(E-cadherin)蛋白和mRNA表达水平明显上调,N钙黏蛋白(N-cadherin)、波形蛋白(Vimentin)、锌指转录因子(Snail)蛋白和mRNA表达水平明显降低(P<0.01)。结论:TEAD2在OSCC中高表达,其高表达与患者不良预后有关,参与OSCC细胞迁移、侵袭及凋亡的调控。

Abstract: Objective:To investigate the expression pattern of transcriptional enhancement-associated domain 2 (TEAD2) and its effect on cell biological behavior in patients with oral squamous cell carcinoma (OSCC). Methods:We selected 97 OSCC and 20 healthy oral mucosa samples. The expression level of TEAD2 in these samples was detected by immunohistochemical (IHC) staining, and the relationships between its expression and clinical parameters and prognosis of OSCC patients were analyzed. Using small interfering RNA (siRNA) to transfect and silence TEAD2 of CAL27 cells and exploring the effects of TEAD2 silencing on the apoptosis, migration and invasion of CAL27 cells by flow cytometry, wound healing assay and transwell invasion assay. The expression of epithelial-mesenchymal transformation related proteins and genes were respectively detected by Western blot and qRT-PCR. Results:IHC results showed that the expression of TEAD2 in OSCC tissues was significantly higher than that of normal oral mucosa (P<0.05). The high expression of TEAD2 was correlated with tumor size, pathological grade, cervical lymph node metastasis. High expression group of TEAD2 had lower overall/disease-free survival rate of patients (P<0.05). After TEAD2 was silenced by siRNA, the apoptotic ability of CAL27 cells was increased, and the invasion and migration ability were decreased significantly (P<0.01). The protein and mRNA expression level of E-cadherin were significantly upregulated, while the expression level of N-cadherin, Vimentin and Snail were significantly decreased after TEAD2 was silenced (P<0.01). Conclusions:Our findings revealed that TEAD2 was overexpressed in OSCC, its high expression was related to the poor prognosis of OSCC patients, and involved in the regulation of OSCC cell migration, invasion and apoptosis.
