负载小檗碱的聚乙烯醇口腔敷料贴片的初步研究

A preliminary study of polyvinyl alcohol oral dressing patches loaded with berberine

摘要: 探究负载小檗碱(BBR)的聚乙烯醇(PVA)贴片作为口腔术后敷料局部应用的可行性。方法将PVA贴片置入不同质量分数(0、2.5%、5%)的BBR水溶液中,待BBR分散均匀后烘干制备成载有不同浓度BBR的PVA贴片材料,对其进行表征检测。采用MTT检测各组贴片的生物相容性,通过抑菌圈法比较各组贴片对牙龈卟啉单胞菌、具核梭杆菌的抑菌作用,通过体外促凝血实验和大鼠断尾模型评估各组贴片的止血性能。结果各组载药贴片材料具有良好的理化性能,能够在体外持续稳定释放BBR,具有良好的生物相容性。与PVA组相比,2.5%BBR/PVA、5%BBR/PVA贴片对两种牙周致病菌均有显著的抑菌作用P<0.001)。体内外止血实验也表明,各组均具有良好的止血作用(P<0.001)。结论负载BBR的PVA贴片材料具有良好的理化性质和生物学特性,且对常见牙周致病菌具有良好的抑菌作用,同时具有良好的止血作用,有望作为一种新型口腔局部抑菌止血材料。

Abstract:

Objective:To investigate the feasibility of a polyvinyl alcohol (PVA) patch loaded with berberine (BBR) for topical application as a postoperative oral dressing. Methods:The PVA patches were placed in aqueous solutions of BBR with different mass fractions (0, 2.5%, 5%), and the PVA patch materials containing different concentrations of BBR were prepared by drying after the BBR was dispersed homogeneously, and they were characterised and tested. The MTT method was used to test the biocompatibility of each group of patches. The antibacterial effects of each group of patch materials on Porphyromonas gingivalis and Fusobacterium nucleatum were compared by the circle of inhibition method. Finally, the haemostatic properties of the patch materials were assessed by in vitro procoagulant assay and rat tail-breaking model. Results:The drug-carrying patch materials in each group had good physicochemical properties and were capable of sustained and stable release of BBR in vitro. The patches in each group showed good biocompatibility. Compared with PVA patches, 2.5% BBR/PVA and 5% BBR/PVA patches showed significant bacteriostatic effects against periodontal pathogens P. gingivalis and F. nucleatumP<0.001. In vivo and vitro haemostatic experiments also showed that all groups had good haemostatic effectsP<0.001. Conclusions:The BBR-loaded PVA patch material has good physicochemical and biological properties, and has good antibacterial effect against common periodontal pathogens and good haemostatic effect, which is expected to be used as a new type of oral topical antibacterial and haemostatic material.
