miR-9对人牙周膜干细胞功能的影响

Effects of miR-9 on the function of human periodontal ligament stem cells

摘要: 目的:探讨miR-9对人牙周膜干细胞(PDLSCs)增殖、迁移和成骨能力的调控作用。方法:分离和培养人PDLSCs,瞬时转染miR-9 inhibitor片段后,使用Brdu法、细胞划痕实验、茜素红染色和Western blot法分别检测PDLSCs的增殖、迁移和成骨能力的改变。结果:瞬时转染miR-9 inhibitor后,PDLSCs的增殖能力减弱,细胞迁移速度变慢,成骨能力受损(P<0.05)。结论:miR-9可调控PDLSCs的增殖、迁移和成骨能力。

Abstract: Objective:To observe the effects of miR-9 on the proliferation, migration and osteogenic differentiation of human periodontal ligament stem cells(PDLSCs) in vitro. Methods: PDLSCs were isolated and cultured in vitro, and were transfected with miR-9 inhibitor. Proliferation of PDLSCs was evaluated by Brdu assay. Migration of PDLSCs was tested by cell wound scratch assay. Osteogenic differentiation of PDLSCs was determined by Alizarin red staining and Western blot. Results: Suppression of the function of miR-9 could reduce proliferation of PDLSCs, lowered the migration speed of PDLSCs, and harmed the osteogenic differentiation of PDLSCs (P<0.05). Conclusions: miR-9 could regulate the proliferation, cell migration and osteogenic differentiation of PDLSCs.
