A methodological study on ergosterol determination from resistant strains of Candida albicans biofilm

Abstract: Objective:To determine Candida albicans biofilm ergosterol with high performance liquid chromatography (HPLC) and gas chromatography-mass spectrometry (GC-MS) methods, and to compare the two methods. Methods:Candida albicans fluconazole-resistant strains were induced by Candida albicans SC5314 with gradually increased fluconazole concentration, and identified by the KONT MIC determination system. Biofilm was formed at 6, 12, 24h. The concentration of ergosterol was determined with HPLC and GC-MS methods. Results:Ergosterol concentration was detected with HPLC, and the concentration of lowest sensitivity is 0.05 mg/L, but it was not detected successfully by GC-MS. Conclusions:HPLC was an ideal way to determine the Candida albicans ergosterol concentration.
