miR-34a通过下调SATB2抑制口腔鳞癌增殖、迁移和侵袭的初步研究

Objective:To investigate expression of miR-34a in oral squamous cell carcinoma (OSCC) tissue samples, and then to study whether it can inhibit the proliferation, migration and invasion of oral squamous cell carcinoma by down-regulating the expression of SATB2. Methods:The expression of miR-34a in OSCC, HN6 cell line was tested by qRT-PCR.We used lentivirus to overexpress SATB2 and constructed miR-34a mimic plasmid, in vitro. Cell proliferation was assessed by CCK-8 experiments and the cell cycles were measured by flow cytometry. Cell migration ability was measured by cell wound healing test and transwell test. Cell invasion ability was measured by trans invasion test and the protein change of cell invasion-related protein MMP-2 and MMP-9 were tested by Western blot. In vivo, the growth of tumor in nude mice was further observed to examine whether miR-34a can affect the growth of transplanted tumors in vivo by modulating the expression of SATB2. Results:The expression of miR-34a in OSCC tissues and cell line are lower than para-carcinoma tissues and oral keratin forms cells (P<0.05). After transfecting miR-34a mimic plasmid, the ability of proliferation, metastasis and invasion of Lv-SATB2-HN6 was significantly reduced. Co-transfection of the miR-34a mimics specifically decreased the expression of the SATB2. Conclusions:MiR-34a can down-regulate SATB2 to inhibit the proliferation, migration and invasion of OSCC.

目的:探究Notch信号通路抑制剂DAPT在体外对人牙周膜间充质干细胞(hPDLSCs)增殖及成骨分化能力的影响。方法:体外培养扩增hPDLSCs,流式细胞术检测间充质干细胞特异性标记物CD73、CD90和造血干细胞特异性标记物CD34、CD45的表达。选取第二代生长良好的hPDLSCs,随机分为3组:空白组使用人间充质干细胞培养基,对照组使用人间充质干细胞成骨诱导培养基,实验组使用含1 μmol/L DAPT的人间充质干细胞成骨诱导培养基进行体外培养。CCK8检测hPDLSCs增殖能力,绘制增殖曲线;实时定量RT-PCR检测碱性磷酸酶(ALP)、Ⅰ型胶原(COL-Ⅰ)、骨钙素(OCN)、骨桥蛋白(OPN)、Runt相关转录因子2(Runx2)等成骨相关基因及Notch信号通路下游基因(Hes-1、Hey-1)和Notch信号通路配体JAG1的表达;茜素红染色显示矿化结节形成情况。结果:原代培养hPDLSCs表面标记物表达CD73(94.49%)、CD90(98.74%)、CD34(2.07%)、CD45(0.62%);实验组在各检测点的细胞增殖能力与对照组之间无明显差别,两组结果无统计学差异(P>0.05);实验组成骨分化相关基因的表达水平升高(P<0.05),Notch信号通路下游基因表达降低,配体表达升高(P<0.05),细胞染色见实验组矿化结节增多增大。结论:DAPT可提高hPDLSCs体外成骨分化能力,但对增殖无明显影响。

Objective:To investigate the effect of BCOR-short on the osteogenic differentiation potential of stem cells from apical papilla (SCAPs). Methods:Retroviral HA-BCOR-short was used to over-express the BCOR-short in SCAPs. Western Blot was used to detect the expression of HA-BCOR-short. Alkaline phosphatase (ALP) activity assay was used to detect the early marker of osteogenic differentiation-ALP activity, Alizarin-red staining and quantitative analysis of calcium were used to investigate the mineralization potential of SCAPs in vitro. Real time RT-PCR was used to detect the osteogenesis related genes expressions bone sialoprotein (BSP), osteocalcin (OCN) and AP2A. Results:Western Blot result showed that HA-BCOR-short could over-express in SCAPs. The over-expression of HA-BCOR-short inhibited ALP activity, mineralization, and the expressions of BSP and OCN in SCAPs. The AP2A expression was repressed in HA-BCOR-short over-expressed SCAPs compared with control group. Conclusions:Our results indicated that over-expression of HA-BCOR-short repressed osteogenic differentiation potential of SCAPs and the expression of AP2A.

Objective: The purpose of this study is to find out the oral microbial community in healthy volunteers by meta-genomic method, and to provide the biological basis for the prognosis and consequences of the treatment of chronic periodontitis. Methods: Total DNA was extracted from healthy volunteers’ saliva samples, supra-gingival and sub-gingival bacterial plaque samples, so as to built genomic libraries. After sequencing and analyzing the results, species were annotated, and their relative abundance was calculated too. Results:The sequencing results showed that there were 15 microbial genuses in saliva samples of healthy volunteers, and there were 20 microbial genuses in samples from healthy volunteers’ supra-gingival and sub-gingival plaques. The mean relative abundance ratio of each healthy volunteers’ microorganism sample group (saliva sample group, supra-gingival sample group and sub-gingival sample group )was significantly different (P<0.05). Conclusions:The meta-genomic method using the second generation high-throughput sequencing technique can obtain the microbial community in the oral samples of healthy volunteers well, and it is capable of helping analyze the microbial community structure in each sample group.

Objective:To explore the proliferation and odonto/osteogenic potential of DPSCs separated from GK rat with hyperglycemia. Methods: The 3-month-old GK rat (one diabetic model rat) and Wistar rat were selected. The DPSCs were isolated and cultured when the blood glucose level had been stable for 1 weeks. MTT assay was applied to estimate the proliferation of the two DPSCs. The odonto/osteogenic markers (OCN, OSX and DMP1) of the two DPSCs were measured by real-time RT-PCR and Western blot. Results: The level of blood glucose in GK rats remained about 20.1~22.3 mmol/L (high glucose) and the level of blood glucose of Wistar rats was 5.8 ~ 6.3 mmol/L (normal glucose level). MTT assay showed the proliferative activity of DPSCs from GK was significantly lower than that of the Wistar rat. Moreover, the expressions of OCN, OSX and DMP1 in DPSCs of GK rats were significantly weaker than those in Wistar rats. Conclusions:The DPSCs from the donor with hyperglycemia or diabetes exhibited the weaker proliferation and odonto/osteogenic potential.

Objective:To explore the difference of somatosensory and jaw function between patients with unilateral temporomandibular joint (TMJ) pain and normal subjects by quantitative sensory test and opening ranges. Methods:20 patients suffering from unilateral TMJ pain and 20 healthy controls were enrolled in this study. Visual analogue scale (VAS),opening ranges (OR) and mechanical pain threshold (MPT), pressure pain threshold (PPT) on both TMJ were measured and compared. Results:VAS of patients were higher than that of controls (P<0.001), but opening ranges were all smaller than controls (P<0.001). Both MPTs and PPTs of pain side of the patients were lower than ipilateral side of controls (P<0.05). MPTs and PPTs were also lower in pain side than those in contralateral side of TMJ in patients (P<0.05). Conclusions:TMJ pain patients demonstrated high sensitivity and limitation of jaw motion .

Objective:To evaluate the clinical efficacy of surgery first or surgery early approach for skeletal Class Ⅲ malocclusion.. Methods: A retrospective analysis of medical records was conducted in 37 patients with skeletal Class Ⅲ malocclusion, who were treated with surgery-first approach in our hospital from July 2010 to June 2017, and preliminary analysis was carried out on the clinical efficacy. Results:All patients were successfully completed the whole procedure with surgery-first approach and all of them were satisfied with the appearance and occlusion, excepting for one case with mild recurrence. The whole period was 6-19 months, with an average of 13.8 months. Conclusions: Computer assisted surgery-first approach can be used in treatment for skeletal Class Ⅲ malocclusion, which not only can shorten the treatment period, but can obtain satisfactory clinical efficacy.

Objective:To comparatively analyze the caries activity among early childhood caries, severe early childhood caries and caries free children. Methods:A total of 289 children aged 3-5 years in three kindergartens in Nanjing were examined for dental caries and caries activity test. The parents were investigated and the results were statistically analyzed. Results:The caries prevalence rate of primary teeth was 47.8%, the prevalence of SECC was 20.4%. Cariostat value was significantly correlated with dmft and dmfs, and had statistical differences between the three groups. Sweet food/drink before sleep and Cariostat value was significantly related to the caries. Conclusions:The cariogenic bacteria activity of early childhood caries can reflect the severity of caries.
