PLGA/胶原双层引导骨再生膜对成骨样细胞增殖的体外研究

In vitro study of the effect of two-layered PLGA/collagen composite membrane for guided bone regeneration on proliferation of osteoblastic cells

摘要: [摘要] 目的:制备一种新型结构的聚乳酸-聚乙醇酸共聚物(poly(lactic-co-glycolic acid),PLGA)/胶原复合引导骨组织再生膜,对其进行表征,并探索其对成骨样细胞体外增殖的影响。方法:使用PLGA及I型胶原制备一种双层结构的引导骨组织再生膜,通过傅里叶变换红外光谱仪、场发射扫描电镜、接触角仪对其表面基团、形貌及亲水性进行测试。将MC3T3-E1成骨细胞系接种到复合膜上,通过流式细胞术、甲基噻唑基四唑(MTT)法检测其细胞增殖,并与对照组进行比较。结果:红外图谱显示胶原被成功结合到PLGA表面,并且在扫描电镜下呈现出特有的纤维网状特征性微结构,同时其表面亲水性大大改善。MTT及细胞周期检测也表明胶原改性后的复合膜上生长的细胞活力及增殖指数明显高于纯PLGA组。结论:本方法制备的PLGA/胶原复合膜很好地组合成了一个功能整体,结合了高分子合成材料和天然材料的优势。

Abstract: [Abstract] Objective:To prepare a novel poly(lactic-co-glycolic acid)(PLGA)/collagen composite membrane for guided bone regeneration, characterize the membrane, and evaluate the effect on proliferation of osteoblastic cells. Methods:PLGA and type I collagen were combined together to prepare a two-layered membrane. The functional groups, surface topography and contact angle of the composite membranes were characterized by attenuated total reflectance Fourier transform infrared spectrometer (ATR-FTIR), field emission scanning electron microscopy (FESEM) and contact angle meter. Osteoblastic cell line MC3T3-E1 was chosen to seed on the membranes. MTT and flow cytometry (FCM) were used to value the cell proliferation. Results:The spectrum of ATR-FTIR showed that collagen layer was successfully grafted onto the PLGA surface. The characteristic microstructure of interlaced fibers of collagen was observed by FESEM. The contact angle of composite membrane was significantly lower than pure PLGA. The results of MTT and FCM demonstrate the cell viability and proliferation index of cells cultured on PLGA/collagen membrane were significantly higher than PLGA. Conclusions: The PLGA/collagen composite membrane prepared in this study combined synthetic and natural polymers into a versatile entirety and adopted the advantages of the both.
