miR-509-3p对头颈鳞状细胞癌细胞CAL27增殖的影响

Effect of miR-509-3p on proliferation of CAL27 cells

摘要: 目的:探讨miR-509-3p对头颈鳞状细胞癌(HNSCC)细胞CAL27增殖的调控作用。方法:实时定量RT-PCR检测头颈鳞癌细胞系HN4、HN6、HN13、CAL27和正常口腔上皮细胞系HOK中miR-509-3p的表达差异。应用miR-509-3p mimic和mimic NC转染CAL27细胞,实时定量RT-PCR检测miR-509-3p表达水平;使用CCK8、克隆形成和Edu等实验技术验证miR-509-3p表达水平变化对CAL27细胞增殖能力的影响;Western blot法检测miR-509-3p表达水平变化对EGFR/PI3K/AKT/MTOR信号通路中各蛋白水平的影响。结果:与HOK细胞株相比,miR-509-3p在头颈鳞癌肿瘤细胞系内表达量明显下调;miR-509-3p mimic显著提高CAL27中miR-509-3p的表达;过表达miR-509-3p能明显抑制CAL27细胞的增殖能力;Western blot结果显示,转染miR-509-3p mimic的CAL27中,表皮生长因子受体(EGFR)、磷酸化表皮生长因子受体(p-EGFR)、磷酸肌醇3激酶(PI3K)、 磷酸化丝氨酸/苏氨酸蛋白激酶(p-AKT)、磷酸化的哺乳动物雷帕霉素靶蛋白(p-MTOR)的表达水平均明显降低。结论:miR-509-3p可能通过调控EGFR/PI3K/AKT/MTOR信号通路影响头颈鳞癌细胞CAL27的增殖能力。

Abstract: Objective: To investigate the regulation of miR-509-3p on the proliferation of head and neck squamous cell carcinoma (HNSCC). Methods:Real-time quantitative RT-PCR was used to detected the expression of miR-509-3p in head and neck squamous cell carcinoma cell lines HN4, HN6, HN13, CAL27 and normal oral epithelial cell line HOK. The CAL27 cells were transfected with miR-509-3p mimic and mimic NC, the expression of miR-509-3p was detected by real-time PCR; Cell proliferation experiments including CCK8, colony formation and EDU analysis have been used to verified the effect of miR-509-3p on proliferation. Results:The expression of miR-509-3p was significantly down-regulated in head and neck squamous cell carcinoma cell lines compared with HOK cells; miR-509-3p mimic can significantly increase the expression of miR-509-3p in CAL27. CCK8, colony formation and EDU analysis have shown that overexpression of miR-509-3p can significantly inhibit the proliferation of CAL27 cells. Western blot results showed that the expression levels of EGFR, p-EGFR, PI3K, p-AKT and p-MTOR were significantly decreased in cells transfected with miR-509-3p mimic. Conclusions:MiR-509-3p can affect the proliferation of CAL27 in HNSCC cell CAL27 by regulating the EGFR/PI3K/AKT/MTOR signal pathway.
