具核梭杆菌感染的巨噬细胞对人舌鳞癌细胞系Cal-27细胞增殖与迁移的影响

Effects of macrophages induced by Fusobacterium nucleatum on proliferation and migration of tongue squamous cell carcinoma Cal-27 cells

摘要: 目的:观察具核梭杆菌(F. nucleatum)诱导的巨噬细胞对人舌鳞癌细胞系Cal-27细胞增殖、迁移的影响。方法:F. nucleatum以感染复数(MOI)100∶1,刺激THP-1细胞来源的巨噬细胞,24 h后收集细胞条件培养液,以THP-1细胞及THP-1细胞来源巨噬细胞的条件培养液为对照。将获得的各组条件培养液与Cal-27细胞共培养,CCK-8法检测各组细胞增殖水平的变化,划痕实验检测Cal-27细胞迁移能力的变化。ELISA检测各组条件培养液中肿瘤坏死因子α(TNF-α)及白细胞介素(IL)-6表达水平。结果:与2个对照组相比,F. nucleatum诱导的巨噬细胞培养液上清使Cal-27细胞增殖减少(P<0.05),相对迁移率增高(P<0.05)。F. nucleatum刺激巨噬细胞后,条件培养液中TNF-α和IL-6水平均较刺激前明显增加(P<0.05)结论:F. nucleatum感染能促进巨噬细胞分泌TNF-α和IL-6,进而可能促进Cal-27细胞迁移,抑制其增殖。

Abstract: Objective:To observe the effects of infection induced by Fusobacterium nucleatum (F. nucleatum) on the proliferation and infiltration of tongue squamous cell carcinoma cell line Cal-27. Methods:Supernatant from THP-1 cells differentiated macrophages stimulated with F. nucleatum (multiplicity of infection was 100:1) for 24 h was collected, and supernatants from unstimulated THP-1 cells differentiated macrophages and THP-1 cells were collected as controls. Cal-27 cells were co-cultured with the above-mentioned supernatants and Cal-27 cells proliferation was detected by CCK-8 method. Migration of Cal-27 cells were evaluated by scratch assay. Expressions of tumor necrosis factor-α (TNF-α) and interleukin (IL)-6 were detected by ELISA. Results: F. nucleatum-induced macrophages conditioned medium reduced the proliferation but promoted the migration rate of Cal-27 cells in comparison with two control groups (P<0.05). The levels of TNF-α and IL-6 in macrophages stimulated with F. nucleatum were significantly higher than those without any stimulations (P<0.05). Conclusions:Stimulation of macrophages with F. nucleatum promoted TNF-α and IL-6 production, which may promote the migration but inhibit the proliferation of Cal-27 cells.
