Objective:To explore the effect of miR-1 on the development of neural crest in zebrafish. Methods:We observed the development of mandible and cranial cartilages at 96 hours post fertilization (hpf) after knockdown the miR-1. Then we carried out the TUNEL and immunofluorescence of phosphorylation of histone H3 (pH3) to study the apoptosis and proliferation of the neural crest cells. Results:In this study, it is demonstrated that knockdown of miR-1 resulted in the impaired craniofacial chondrogenesis. We observed reduced lower jaw, abnormal mandibular and ceratohyal cartilage. We also found knockdown of miR-1 promoted the cell death and suppressed the proliferation during the development of neural crest cells. Conclusions:These results indicate an important role for miR-1 in neural crest cells development and suggest involvement of miR-1 in neural crest cell viability.

Objective: We designed a novel strategy for enamel remineralization, including an amelogenin-like oligopeptide as templates for controlling the nucleation and growth of HA crystals and synthesized calcium- and phosphate-loaded temperature sensitive liposomes supplying calcium and phosphate ion, to induce HA crystals and promote biomimetic mineralization. Methods:A novel polyanion amelogenin-inspired oligropeptide (Gln-Pro-Ala)4-Thr-Lys-Arg-Glu-Glu-Val-Asp) was synthesized by standard solid-phase, and induced self-assembly with CaCl2 solution. Secondly, the calcium/phosphate phosphate loaded liposomes were prepared using the interdigitation-fusion method. The liposomes were composed of 1,2-bis(palmitoyl)-sn-glycero-3-phosphocholine and 1,2-bis(myristoyl)-sn-glycero-3-phosphocholine. Lastly, the experimental groups treated with mixture solution of liposomes and polyanion peptide. But the control groups were treated with the liposomes solution. These two groups were characterized by scanning electron microscope(SEM), Fourier transform infrared spectroscopy(FTIR) and X-ray Diffraction(XRD). Results:The experimental groups showed that the rod-like crystals almost fully covered the etched enamel surface, and distributed evenly to make the surface smooth. In contrast, the control groups showed that there was only small amount of HA crystal deposition, and the precipitate distributed randomly. Conclusions:The self-assembly amelogenin-inspired oligopeptide polyanion peptide combining with temperature sensitive Ca2+ and PO43- loaded liposomes can effectively induce biomimetic mineralization of enamel.

[Abstract]Objective: Adenosine deaminase 1 acting on dsRNA ( ADAR1) may be involved in the regulation of microRNA biogenesis and implicated in tumor development. Here we aimed to examine the of ADAR1 in oral squamous cell carcinoma (OSCC) and its influence on biological behavior of OSCC. Methods: The of ADAR1 in OSCC cell lines was tested by RT-qPCR and Western Blot. The of ADAR1 gene in OSCC cell lines was knocked down by siADAR1. Wound healing and Transwell test were used to detect migration and invasion of OSCC. RT-qPCR and Western Blot were used to detect epithelial-mesenchymal transition(EMT) markers Vimentin and E-cadherin, stemness markers Sox2 and Oct4, and OSCC-associated onco-microRNAs(miR-21-3p、miR-18a-3p、miR-210-3p、miR-155-5p、miR-181a-3p、miR-19a-3p). Results: Higher ADAR1 was expressed in OSCC compared to normal oral mucosa. After ADAR1 knocked down, not only the migration and invasion of OSCC were significantly inhibited (P<0.05), but also EMT and stemness markers such as Vimentin, Sox2, and Oct4 decreased. Of note, OSCC-associated onco-microRNAs (miR-21-3p、miR-18a-3p、miR-210-3p、miR-155-5p、miR-181a-3p、miR-19a-3p) also decreased with ADAR1 gene knock down. Conclusions: ADAR1 is closely related with the biological behavior of OSCC. We speculate that ADAR1 may have impact on the biological behavior of OSCC by promoting mature microRNA processing.

Objective: Investigate the role of miR-223 on the of RANKL in HPDLFs. Methods:The HPDLFs over expressed with miR-223 was constructed, the of RANKL and OPG were detected by western blot and real time PCR, and the change of RANKL/OPG ratio was calculated. Results: The RANKL was down-regulated in the HPDLFs with miR-223 over expressed and the RANKL/OPG ratio was decreased. Conclusions: MiR-223 could disrupt the homeostasis of RANKL/OPG, destroy the microenvironment of osteoclast differentiation. MiR-223 may play a regulatory role in osteoclast differentiation .

Objective:To research the effect of xeno-ADM (xeno-acellular dermal matrix) on vasculogenesis of human umbilical vein endothelial cell(HUVEC).Methods:Porcine acellular dermal matrix (P-ADM) and bovine acellular dermal matrix (B-ADM) were co-cultured with HUVEC, using blank and Medical collagen membrane(Bio-Gide) as control. The concentration of vascular endothelial growth factor (VEGF) collected from different groups at 4, 6, 12, 24, 48, 96 h was determined by ELISA test. Meanwhile, the number of cellular casts were counted by observing HUVEC infected by GFP lentivirus. Results:The VEGF concentration of xeno-ADM groups were higher than that of the control groups(P<0.05)after 24 h, and P-ADM was more than that of the control groups after 48 h(P<0.05).The number of cellular casts of P-ADM group was about as twice as that of the control group after 24 h. The B-ADM group improved less than the P-ADM group. No cellular cast was found in the control group..Conclusions:Xeno-ADM has better effect on vasculogenesis than traditional medical collagen membrane, which suggesting that it has the potential to be the next generation of surgical material applying to mucogingival therapy.

Objective:Studies on the effect of cryopreservation on the biological characteristics of dentine and dental pulp cells in vitro. Methods:A total of 40 fresh teeth (the first premolar or second premolar) were collected and respectively experiment with dentine mechanical morphological analysis and dental pulp cells growth experiments. At the experiment of dentine mechanical morphological analysis, 20 fresh extracted human premolars were selected,prepared into dentin discs,and randomly divided into 2 groups (group A:fresh teeth group,group B:programmed cooling group), which were measured for compression strength and scanning electron microscope analysis. Another 20 teeth, using improved tissue block method of separation of dental pulp cells, the original generation of cryopreserved after recovery were observed before and after analysis of cryopreserved cells morphological and growth characteristics. Results:There were no obvious differences between groups with cryopreservation in growth characteristic of dentine and dental pulp cells. Conclusions:Tooth pulp tissue can keep good biological characteristics with cryopreservation.

Early orthodontic intervention in pediatric dentistry also has showed a rapid development and achieved a major breakthrough. The dentists use all kinds of methods, including preventing the occurrence of malocclusion and blocking progress of malocclusion and other measures, to create a healthy oral environment and reduce the difficulty of comprehensive orthodontic treatment in permanent dentition. We believe that the application of occlusive guidance and functional treatment in early orthodontic intervention of preadolescent malocclusions can achieve good maxillofacial development and occlusal construction, so as to promote physical and mental healthy development of children. This study reviewed the development and clinical application of occlusive guidance and functional treatment, combined with the causes of malocclusion and the characteristics of preadolescent growth, in order to provide guidance for early orthodontic intervention of preadolescent malocclusion.

Congenital cleft lip and palate(CLP)is the most common one of the birth defects, which has a serious impact on the oral function,facial morphology and mental health.It is a constant pursuit of scholars to restore the aesthetic and functional defects in patients with CLP.Clinically,Operaters should make surgery programs and predict postoperative facial morphology based on accurate quantitative measurement of maxillofacial area.Commonly used two-dimensional measurements are photo measurement and X-ray measurement,both are simple to operate and practical,low cost,but not accurate enough.Three-dimensional measurements such as computer tomography, stereoscopic photography,laser scanning can analyze the facial shape objectively and quantitatively,These three-dimensional measurements have broad application prospects with high accuracy,efficiency and convenience.Moreover,With the development of dynamic three-dimensional imaging technology,the dynamic 3D image capture technology is applied in dynamic analysis of maxillofacial region,and is expected to play an important role in the measurement of patients with CLP.
